<p>In a study published this week in the Proceedings of the National Academy of Sciences, researchers from the University of Chicago and the Andalusian Center for Developmental Biology in Spain used genetic editing tools to show how a gene that controls the growth of bones at the terminal end of fish fins play the same role in forming fingers and toes in four-legged creatures. The same gene also controls this process both in paired fins, which are the progenitors to limbs, and the single, unpaired dorsal fin common to all fish that evolved before paired fins. This suggests that the last common ancestor between ray- and lobe-finned fish nearly 500 million years ago already had the genetic toolkit to shape their appendages, shared to this day by fish and four-legged vertebrates.</p>
<p>?There?s this deep homology or similarity between fins and limbs, something ancient in structures that really don?t look alike,? said Neil Shubin, PhD, the Robert R. Bensley Distinguished Service Professor of Anatomy at UChicago and co-author of the new study. ?We?re showing a deeply conserved, deeply ancient and preserved gene function that?s been around for hundreds of millions of years in vastly different structures. So, the molecular toolkit is ancient, and it does the same thing in different kinds of animals.?</p>
<p>The study is a continuation of a collaboration between Shubin and the late José Luis Gómez-Skarmeta from Spain, which began at the UChicago-affiliated Marine Biological Laboratory in Wood Hole, Massachusetts. In 2018, they published a study that tracked the expression of the Sonic hedgehog (Shh) gene, which is widely used in a variety of basic biological functions, but especially important in the formation of limbs. In that study, they identified a combination of genetic enhancers that controlled development of limbs in mice but affect dorsal and paired fins in fish separately.</p>
<p>In the new research, Shubin, Gómez-Skarmeta, and Joaquin Letelier, also of the Andalusian Center, focused on another gene that works with Shh called gli3. This gene is already known to pattern the digits in a limb, helping determine the identity of each from thumb to pinky. Humans who have a mutation in gli3 often grow extra digits, what?s called polydactyly, and mice who have gli3 knocked out in the lab also grow extra digits. The researchers wanted to see if gli3 worked the same way in fish, so they used CRISPR gene-editing tools to knock it out in the medaka, a small, popular aquarium fish also known as a Japanese rice fish. The gli3-knockout fish also developed the fish version of polydactyly, with multiple radial bones at the base of the fin and more fin rays. Interestingly, this happened in the paired pectoral and pelvic fins (the fish analog to limbs) and the unpaired, evolutionarily older, single dorsal fin as well.</p>
<p>Research in mice and chicken embryos shows that gli3 is involved in the process of cell proliferation, which makes sense for its role in polydactyly; If knocking out the gene causes more cells to grow in the appendage, it will also lead to more digits. Further genetic analysis in the fish showed that gli3 was also involved in cell proliferation in the fins, which suggests strong evolutionary similarities across species. As later terrestrial animals developed more advanced limbs, gli3 took on a more specialized role to control their shape and patterning.</p>
<p>?The speculation is that you have a primitive function of gli3 present in all vertebrate appendages ever since about 500 million years ago, and that was to promote proliferation, or the number of cells and hence the number of bones in the terminal end,? Shubin said. ?When paired fins arose it was already there, so gli3 was co-opted and gained a new role, which is anterior/posterior patterning.?</p>
<p>Shubin says powerful new genetic tools like CRISPR and sequencing continue to allow scientists to track down more fascinating clues like this about the deep, shared history of all vertebrates.</p>

<li class="css-4cm648"><span>The six best group runners-up from the World Cup qualifiers (including Sweden, by all accounts) are seeded and drawn against the unseeded teams.  The seeded teams can play at home in the semifinals.  Venue in the finals is decided via a draw.</span></li>
<li class="css-4cm648"><span>The playoff matches are played as single meetings.  The semifinals are played March 24-25, the finals March 28-29.</span></li>

<p><strong>Recently, Nordic Clinic organised a joint webinar together with Food Pharmacy on the theme autoimmunity. We are so happy that a big bunch of Food Pharmacy?s followers signed up for the webinar. The great interest in the topic is most likely a consequence of the growing group of people diagnosed with an autoimmune disease. </strong></p>
<p>As we discussed in the webinar, autoimmune disease is not pretty, but there are many factors that may help those with an autoimmune diagnosis. Autoimmune diseases vary widely in their presentation and symptomatology due to different target organs. A few autoimmune conditions and the target organs/systems include:</p>
<ul>
<li>Hashimoto?s Thyroiditis (Thyroid)</li>
<li>Type 1 Diabetes (Pancreas)</li>
<li>Rheumatoid Arthritis (Joints)</li>
<li>Multiple Sclerosis (Central Nervous System)</li>
<li>Inflammatory Bowel Disease (GI Tract)</li>
</ul>
<p>Though these conditions lead to a range of symptoms, the underlying driver is an immune system that turns its attention to our own tissues. Diet may be an important factor one can change to help manage an autoimmune condition. The underlying mechanism involves changes in the gut microbiome, intestinal permeability, or removal of dietary antigens that resemble human tissue.</p>
<p>A recent review highlights the complexities in the relationship between diet and autoimmune conditions.<sup>1</sup></p>
<h4>Diet As Poison</h4>
<p>Various aspects of the modern diet may play a role in driving chronic inflammation. Gluten has inflammatory properties in certain individuals, particularly those with celiac disease.<sup>2</sup> In some individuals, the gliadin component of gluten has been shown to activate immune receptors in the gut called TL4. The microbiome likely plays a role here as well, so you want to look after those little critters in your gut. </p>
<p>High intake of long chain saturated fatty acids found in meat and dairy products, coconut oil and some vegetable oils, is also believed to induce inflammation via a mechanism involving TLR4.<sup>3</sup> Furthermore, it appears to increase lipopolysaccharide (toxins released from bacteria) abundance in the gut and translocation in to the body.<sup>4</sup></p>
<p>Other nutritional factors within the diet may also play a role in inducing inflammation and promoting autoimmune disease. This includes dairy and various food additives such as dietary emulsifiers.</p>
<h4>Diet As Health Promoting</h4>
<p>It has become popular to demonise diets that include any of the above. But the truth is, many diets that include these components benefit autoimmune disease. This is because other dietary components may have a calming effect on the immune system.</p>
<p>For example, Mediterranean and vegetarian diets contain gluten and have been shown to benefit autoimmune conditions.<sup>1</sup> This may be due to their high content of fiber and polyphenols from fruits and vegetables. </p>
<p>Alternatively, the Paleolithic diet may benefit those with Multiple Sclerosis, despite being high in long chain saturated fatty acids.<sup>5</sup> Furthermore, it seems to have a beneficial effect on intestinal permeability and inflammation. This may be due, in part, to a higher intake of whole, unprocessed foods and elimination of processed foods.</p>
<p>Ultimately, a personalised dietary approach to treating autoimmune disease should look at both the beneficial and detrimental components of what we eat.</p>
<h4>Using Diet To Target Autoimmunity </h4>
<p>Diet is an attractive target for the management of autoimmune conditions. Ideally, patients should work with healthcare professionals, nutritional therapists and dietitians to identify the appropriate diet. Individual factors such as genetics, the microbiome, and disease presentation/affected target organ will drive this personalised approach.</p>
<p>Finally, a delicate balance between the elimination of detrimental foods and inclusion of beneficial foods is important. Tailoring the approach using an exclusion diet (Mediterranean, AIP, etc.) followed by re-challenge is key. This approach helps individuals with autoimmune disease find the diet that?s right for their situation.  </p>

<p>Earth?s planetary core is thankfully intact. But in space, similar cores can collide with other objects, causing the crystalline materials of the core to deform rapidly. Some asteroids in our solar system are massive iron objects that scientists suspect are the remnants of planetary cores after catastrophic impacts.</p>
<p>Measuring what happens during the collision of celestial bodies or at the Earth?s core is obviously not very practical. As such, much of our understanding of planetary cores is based on experimental studies of metals at less extreme temperatures and pressures. But researchers at the Department of Energy?s SLAC National Accelerator Laboratory have now observed for the first time how iron?s atomic structure deforms to accommodate the stress from the pressures and temperatures that occur just outside of the inner core.</p>
<p>The results appear in Physical Review Letters, where they have been highlighted as an Editor?s Suggestion.</p>
<p><strong>Coping with stress</strong></p>
<p>Most of the iron you encounter in your everyday life has its atoms arranged in nanoscopic cubes, with an iron atom at each corner and one in the center. If you squeeze these cubes by applying extremely high pressures, they rearrange into hexagonal prisms, which allow the atoms to pack in more tightly.</p>
<p>The group at SLAC wanted to see what would happen if you kept applying pressure to that hexagonal arrangement to mimic what happens to iron at the Earth?s core or during atmospheric reentry from space. ?We didn?t quite make inner core conditions,? says co-author Arianna Gleason, a scientist in the High-Energy Density Science (HEDS) Division at SLAC. ?But we achieved the conditions of the outer core of the planet, which is really remarkable.?</p>
<p>No one had ever directly observed iron?s response to stress under such high temperatures and pressures before, so the researchers didn?t know how it would respond. ?As we continue to push it, the iron doesn?t know what to do with this extra stress,? says Gleason. ?And it needs to relieve that stress, so it tries to find the most efficient mechanism to do that.?</p>
<p>The coping mechanism iron uses to deal with that extra stress is called ?twinning.? The arrangement of atoms shunts to the side, rotating all the hexagonal prisms by nearly 90 degrees. Twinning is a common pressure response in metals and minerals ? quartz, calcite, titanium and zirconium all undergo twinning.</p>
<p>?Twinning allows iron to be incredibly strong ? stronger than we first thought ? before it starts to flow plastically on much longer time scales,? Gleason said.</p>
<p><strong>A tale of two lasers</strong></p>
<p>Reaching these extreme conditions required two types of lasers. The first was an optical laser, which generated a shock wave that subjected the iron sample to extremely high temperatures and pressures. The second was SLAC?s Linac Coherent Light Source (LCLS) X-ray free-electron laser, which allowed the researchers to observe the iron on an atomic level. ?At the time, LCLS was the only facility in the world where you could do that,? says lead author Sébastien Merkel of the University of Lille in France. ?It?s been a door opener for other similar facilities in the world.?</p>
<p>The team fired both lasers at a tiny sample of iron about the width of a human hair, hitting the iron with a shock wave of heat and pressure. ?The control room is just above the experimental room,? Merkel says. ?When you trigger the discharge, you hear a loud pop.?</p>
<p>As the shock wave hit the iron, researchers used the X-ray laser to observe how the shock changed the arrangement of the iron atoms. ?We were able to make a measurement in a billionth of a second,? Gleason says. ?Freezing the atoms where they are in that nanosecond is really exciting.?</p>
<p>The researchers collected these images and assembled them into a flipbook that showed iron deforming. Before the experiment was complete, they didn?t know if iron would respond too fast for them to measure or too slow for them to ever see. ?The fact that the twinning happens on the time scale that we can measure it as an important result in itself,? Merkel says.</p>

<p>This experiment serves as a bookend for understanding the behavior of iron. Scientists had gathered experimental data on the structure of iron at lower temperatures and pressures and used it to model how iron would behave at extremely high temperatures and pressures, but no one had ever experimentally tested those models.</p>
<p>?Now we can give a thumbs up, thumbs down on some of the physics models for really fundamental deformation mechanisms,? Gleason says. ?That helps to build up some of the predictive capability we?re lacking for modeling how materials respond at extreme conditions.?</p>
<p>The study provides exciting insights into the structural properties of iron at extremely high temperatures and pressures. But the results are also a promising indicator that these methods could help scientists understand how other materials behave at extreme conditions, too.</p>
<p>?The future is bright now that we?ve developed a way to make these measurements,? Gleason says. ?The recent X-ray undulator upgrade as part of the LCLS-II project allows higher X-ray energies ? enabling studies on thicker alloys and materials that have lower symmetry and more complex X-ray fingerprints.?</p>
<p>The upgrade will also enable researchers to observe larger samples, which will give them a more comprehensive view of iron?s atomic behavior and improve their statistics. Plus, ?we?re going to get more powerful optical lasers with the approval to proceed with a new flagship petawatt laser facility, known as MEC-U,? says Gleason. ?That?ll make future work even more exciting because we?ll be able to get to the Earth?s inner core conditions without any problem.?</p>

<p>?The future is bright now that we?ve developed a way to make these measurements,? Gleason says. ?The recent X-ray undulator upgrade as part of the LCLS-II project allows higher X-ray energies ? enabling studies on thicker alloys and materials that have lower symmetry and more complex X-ray fingerprints.?</p>
<p>The upgrade will also enable researchers to observe larger samples, which will give them a more comprehensive view of iron?s atomic behavior and improve their statistics. Plus, ?we?re going to get more powerful optical lasers with the approval to proceed with a new flagship petawatt laser facility, known as MEC-U,? says Gleason. ?That?ll make future work even more exciting because we?ll be able to get to the Earth?s inner core conditions without any problem.?</p>

Dus ging het beest de schuur in, maar dan aan een haak of in een kuip met pekel. Om de winter mee door te raken als er minder groenten waren.</p>
<p>Hetzelfde gebeurde met het konijn. Konijnen zijn voor de wetenschap <em>lagomorfen</em> (de orde van konijnen en hazen) en dus géén knaagdieren! Onze konijnen (<em>Oryctolagus cuniculus L</em>) zijn destijds vanuit Spanje en de Maghreb in noordelijk Europa geïntroduceerd. Daarvoor leefden ze alleen in die gebieden. Ze waren aantrekkelijk vanwege hun handige formaat en bescheiden noden in het neerhof. Konijnen werden gevoerd met gras. Gras uit de bermkant. Iedereen kon erbij en dat gras diende ook nergens anders voor. Vaak was het een taak van de kinderen om elke dag gras te snijden en naar het konijnenhok te brengen.</p>

<p><strong>Konijn met keutels</strong><br />Hak het dier in handige stukken. De billen gaan apart, de voorpoten ook en dan is er de heerlijke rug. Niertjes en lever worden even opzij gehouden.</p>
<p>Wentel de stukken konijn door de bloem. Die zal straks de saus binden. In een grote cocotte laat je nu de stukken kleuren in boter of smout. Om en om draaien tot alles wat bruin ziet en dan opzij houden. Dan doe je ajuinen en sjalotten in het vet, in niet te kleine stukken gesneden. Hierop ook nog wat bloem en kruiden: peper en zout, peterseliestelen. Salie is ook niet slecht bij dit witte vlees, de durvers gaan voor steranijs. Leg het vlees erbij en giet hier genoeg bier over tot het bijna onder staat. Voor dergelijke bereidingen kies je best voor bier met weinig hop, zoals rodenbach, geuze. Strooi er nu nog met gulle hand wat gedroogde pruimen over. Hoe die ooit in het recept zijn terechtgekomen is een even grote vraag als de amandelen bij de forel. Maar het hoort zo. Mijn moeder hield er niet van, ze verving de pruimen door rozijnen en krenten. ?Kijk?, zei ze dan, ?konijn met keutels!?</p>
<p><strong>Niet zoals bij de Griek</strong><br />Indien u kriekbier gebruikt, neem dan geen pruimen, maar voeg op het einde een pot krieken op lichte siroop toe. Proef na een tijdje de saus. Misschien moet er een toets zuur bij (scheutje azijn). Zoet zou het genoeg moeten zijn met de pruimen. Laat heel rustig sudderen, een klein uurtje, denk ik. Schep nu en dan om. Vijftien minuten voor alles klaar is, voegt u er de lever en de niertjes bij. Die lever is ware godenspijs!</p>
<p>Laat het geheel vooral niet te lang stoven. Ik heb bij Griekse restaurants <em>Lapin à la grecque</em> gegeten dat echt niet meer te kauwen viel, alleen nog draden en vezels! Dat heeft mij lang van bij de Griek weggehouden. Zoals met alle volkse streekgerechten: deze bereiding is de volgende dag nog lekkerder. Smakelijk.</p>

<p>?The key is to be transparent in messaging. Our model was developed to address food incidents and it highlights strategies to use to communicate effectively with the public. The same ideas make sense in a COVID-19 situation.?</p>
<p>The Flinders researchers? model identifies10 strategies, including transparency; development of protocols and procedures; credibility; proactivity; putting the public first; collaborating with stakeholders; consistency; education of stakeholders and the public; building your reputation; and keeping your promises.</p>
<p>The model has been has presented to key government bodies including SA Health and Food Standards Australia New Zealand, and the original work was then replicated in Ireland.</p>
<p>?We have found that in times of crisis through a major food incident ? when the public may doubt who they trust ? it is imperative that the messages of the food regulators and government authorities are trusted so that the public can act accordingly in line with recommendations.</p>
<p>?Therefore, the model we created focuses on how authorities can best communicate key messages to the public ? which in a pandemic involves key behaviours like social distancing.?</p>
<p>While pandemic management differs from a food incident ? as the responsibility to act is with the public rather than identifiable regulatory bodies, and governments must weigh competing risks in creating policy ? the Flinders researchers conclude that many of the strategies identified in their food trust model could be successfully applied to the maintenance of trust in public health officials prior to, during, and after pandemics.</p>
<p>?The ultimate goal is to maximise trust between the public and governments in Australia, to support public adherence of public health recommendations in response to COVID-19, such as social distancing and isolation,? says Dr Wilson.</p>
<p>?Ideally, we would like to test the application of this model in the COVID-19 pandemic context, and then roll it out for use by state and federal governments across Australia. We currently have a grant application under review to ideally help us to do this.?</p>
