Haste makes waste, as the saying goes. Such a maxim may be especially true of batteries, thanks to a new study that seeks to identify the reasons that cause the performance of fast charged lithium-ion batteries to degrade in electric vehicles.

In new research from the U.S. Department of Energy?s (DOE) Argonne National Laboratory, scientists have found interesting chemical behavior of one of the battery?s two terminals as the battery is charged and discharged.

Lithium-ion batteries contain both a positively charged cathode and a negatively charged anode, which are separated by a material called an electrolyte that moves lithium ions between them. The anode in these batteries is typically made out of graphite ? the same material found in many pencils. In lithium-ion batteries, however, the graphite is assembled out of small particles. Inside these particles, the lithium ions can insert themselves in a process called intercalation. When intercalation happens properly, the battery can successfully charge and discharge.

When a battery is charged too quickly, however, intercalation becomes a trickier business. Instead of smoothly getting into the graphite, the lithium ions tend to aggregate on top of the anode?s surface, resulting in a ?plating? effect that can cause terminal damage ? no pun intended ? to a battery.

?Plating is one of the main causes of impaired battery performance during fast charging,? said Argonne battery scientist Daniel Abraham, an author of the study. ?As we charged the battery quickly, we found that in addition to the plating on the anode surface there was a build up of reaction products inside the electrode pores.? As a result, the anode itself undergoes some degree of irreversible expansion, impairing battery performance.

Using a technique called scanning electron nanodiffraction, Abraham and his colleagues from the University of Illinois Urbana-Champaign observed another notable change to the graphite particles. At the atomic level, the lattice of graphite atoms at the particle edges becomes distorted because of the repeated fast charging, hindering the intercalation process. ?Basically, what we see is that the atomic network in the graphite becomes warped, and this prevents lithium ions from finding their ?home? inside the particles ? instead, they plate on the particles,? he said.

?The faster we charge our battery, the more atomically disordered the anode will become, which will ultimately prevent the lithium ions from being able to move back and forth,? Abraham said. ?The key is to find ways to either prevent this loss of organization or to somehow modify the graphite particles so that the lithium ions can intercalate more efficiently.?

Although pharmacology is a fact of life is, little of the research in that scientific discipline penetrates the public media. Yes, when a major pharmaceutical company has developed a promising new drug (a Covid pill, for example), it is highly praised. After all, successful sales start with good marketing. But research articles that appear in journals such as Pharmacological Reviews The blogs, newspapers, radio and TV usually do not make it.

There is at least one clear reason for this: they are incomprehensible to the uninitiated reader. This also applies to the article ‘Pharmacological Modulation of Immune Responses by Nutritional Components’ that appeared in the above-mentioned journal. The four authors, Marthe T. van Daal, Gert Folkerts, Johan Garssen and Saskia Braber, are affiliated with the Department of Pharmaceutical Sciences of Utrecht University (UU) and conduct fundamental research into the effect of medicines and other substances at the molecular and cellular level.

Use
That their work is now once again receiving wider attention, including through a items in Algemeen Dagblad, is not only justified, but in this case also really newsworthy. In a few words: the researchers are building a (biological) logical bridge between pharmacology and nutrition. That bridge was already across the gap between the two disciplines, but strangely enough, it was rarely walked on.

Assistant professor Saskia Braber agrees that the collaboration between the two disciplines was not very close, but sees a convergence. ?It was always two niches. Pharmacologists didn’t look at food. Medical students used to receive only a few hours of nutrition courses in the entire programme, I recently heard. At the University of Wageningen you have a nutrition department and we have pharmacy in Utrecht, but it is only in recent years that we are working together more and more.?

Research into how certain substances, medicines or foods, have an effect on a cellular level has recently become a lot more accurate

Collaboration between different disciplines is essential. This is organized, for example, on the so-called research hub Future Food Utrecht, the initiative of Utrecht University that as mission statement has: ‘Planetary Health: how do we future-proof our food system?’ In addition, the university also participates in a knowledge center in the field of carbohydrates, the Carbohydrate Competence Center, a private-public partnership with, among others, the universities of Groningen and Wageningen and various companies.

receptors
There is a reason for the convergence between the disciplines. Research into how certain substances, medicines or foods, have an effect on a cellular level has recently become a lot more accurate. In their article, the Utrecht researchers describe the molecular mechanisms that trigger certain nutrients after they act on receptors in the cells, as it is called. That is very similar to what happens with certain medicines. QEDfood as medicine.

?Notably, there is currently very little clinical and reliable research available on the immunomodulatory effects of food components via receptor signalling,? the researchers write. They reviewed the relevant literature and presented a list of four groups of known ‘pharmacological’ receptors, through which food components and their metabolites also have an effect on the immune system.

That list is ?a tool to better understand the pro-inflammatory and/or anti-inflammatory properties of these dietary components, which could aid efforts to prevent and treat NCDs.? ‘Noncommunicable diseases’ are non-contagious ‘wealth diseases’, such as type 2 diabetes, cardiovascular diseases and some cancers.

Spikes
We live in the age of biology, in the age of proteins, in the days of the… spikes, always new molecular nails on the corona mutants. The director of Pfizer Netherlands is happy to meet Beau and his colleagues with a smile to explain how clever they are in his lab. That mechanism, of complex proteins that open the doors to cells, is the work of similar receptors, described by Braber and colleagues in relation to certain food components.

Braber and her team research a large number of foods themselves. ?We look at the effect of substances such as complex carbohydrates, polyunsaturated fatty acids, probiotics and prebiotics, you name it. We look at what effect they have on the cells of the immune system, what effect on the cells in the gut, on pathogenic bacteria and viruses. What diseases can they influence and even prevent? We are conducting research into how we can make the intestinal barrier stronger so that fewer pathogens can pass through. We specialize in that. We test which medicines or nutrients can restore the intestinal wall.?

breast milk
In addition to having a prebiotic effect in the gut, some oligosaccharides also act directly on immune cells. That is a fairly new branch of research for us, says Braber. ?We have shown that certain oligosaccharides are not only processed by bacteria, but also act directly on immune cells and cells of the intestinal wall, thereby strengthening the intestinal barrier. We are conducting a study in which we are looking at the effect nutrients have on the lungs, what the interaction is between the gut and the lungs. We are even investigating whether we can administer nutrients with nasal sprays.?

Braber himself studies the oligosaccharides that occur in breast milk. The idea behind this is that they play such an important role in building a child’s immune system that they should also be able to work later in life.

?These sugars can do much more than we think, promote good bacteria, inhibit bad ones and, apart from that, stimulate the immune system. I think there is still a lot to learn about the well-known foods, the sugars, the fatty acids and the proteins in the future. The advantage of collaborating with the business community is that you are working on something that can be produced at a later date and which may come on the market for the benefit of patients.?

This proof of principle study, published in Nature Communications today (Friday 3rd December), demonstrates how the technology could be used to improve animal welfare in scientific research and perhaps also agriculture.

In scientific research and also farming, there is often a need for either male or female animals. For example, laboratory research into male or female reproduction requires only animals of the sex being studied. And in farming, only female animals are required for egg production and in dairy herds. This means it is common practice for animals of the unrequired sex to be culled after birth.

The researchers? new method uses a two-part genetic system to inactivate embryos shortly after fertilisation, allowing only the desired sex to develop. Such a genetically-based method to control the sex of offspring could drastically reduce culling in both industries.

The embryo selection is based on the fact that there are two elements of CRISPR-Cas9 ? the Cas9 enzyme that cuts the DNA, allowing scientists to alter specific regions, and the guide RNA which carries the Cas9 to the right location on the genome. The team placed one element of the system on the father?s X or Y chromosome, meaning that it will only be inherited by female or male embryos respectively. The other element is contributed by the mother, and is inherited by all embryos.

They targeted the Top1 gene, which is essential to DNA replication and repair. When an embryo formed from a sperm and egg, each containing one half of CRISPR-Cas9, the gene-editing was triggered in the embryo and it was not able to develop beyond a very early stage of around 16 to 32 cells.

Using this method, the researchers were able to control, with 100% effect, the sex of a litter. To produce a male-only litter, the researchers edited the father?s X chromosome, meaning only females inherited the deleterious mutation, and for a female-only litter, they edited the Y chromosome.

Surprisingly, the method did not lead to a 50% decrease in the number of offspring produced, rather the litter sizes were between 61% ? 72% of the control litters. The researchers suggest this is because animals such as mice produce more eggs than required during each ovarian cycle, allowing for a proportion of them to be lost during early development without reducing litter size. This means that in situations where one sex is needed, fewer breeding animals will be required in order to produce the same number of the desired sex of offspring.

As the Top1 gene is well conserved across mammals, these results may also be applicable to other animals.

Charlotte Douglas, first author and former PhD student and postdoctoral scientist at the Crick, says: ?This method works as we split the genome editing process in half, between a male and female, and it is only when the two halves meet in an embryo through breeding, that it is activated. Embryos with both halves cannot develop beyond very early cell stages.

?We?ve also shown this process works successfully in different combinations ? introducing either the Cas9 or the guide RNA elements on to the mother?s or father?s chromosomes.?

As the offspring which survive only contain half of the CRISPR-Cas9 elements within their genome, this acts as a control preventing the sex-selection being passed down to further generations, unless they are selectively bred with an individual of the opposite sex containing the other half. This is different to genetic engineering through ?gene-drive? methods, which seek to spread a genetic mutation widely amongst a population.

There are also no harmful effects of the gene edit in the surviving offspring.

James Turner, author and group leader of the Sex Chromosome Biology Laboratory at the Crick says: ?This work could have immediate and valuable impact in scientific laboratories, as we?ve shown how it is safe and effective in mice, a common mammal used in medical and scientific research. While a lot of research needs both sexes, there are areas of study where only one is needed. For example, when studying the reproductive system, sex-specific diseases, or certain hormones.?

Peter Ellis, author and senior lecturer in molecular genetics and reproduction at University of Kent, says: ?The implications of this work are potentially far-reaching when it comes to improving animal welfare, but should be considered at ethical and regulatory levels.

?In particular, before any potential use in agriculture, there would need to be extensive public conversation and debate, as well as changes to legislation. On the scientific side, there is also much work to be done over a number of years. Further research is needed, first to develop the particular gene editing toolkits for different species, and then to check they are safe and effective.?

Thirty years after winning his first Oscar for one of the most iconic characters in film history, the Hannibal Lecter of “The Silence of the Lambs”, Anthony Hopkins could repeat his statuette with “The Father” in a radically opposite and fragile register of a man suffering from senile dementia.

In real life, the Welsh-born actor displays a friendly demeanor and excellent humor that contrasts with the turbulence of both characters: he tells anecdotes, recites poems and shoves his audience in his pocket, including Olivia Colman, his co-star and daughter in fiction, turned into one more listener in a telematic interview with various international media.

“Life is absurd, it’s ridiculous, we take everything too seriously,” says 83-year-old Hopkins. “I try to cultivate the art of indifference, not in a sense of coldness, but the things that a few years ago I thought important now do not seem so to me, we are all going to die and that is the blackest and funniest joke.”

“The father”, premiered in Spain on December 23 and now expanding its presence in theaters for the Oscars – whose nominations are announced today – is the debut of the French writer and playwright Florian Zeller. His great discovery is that he places the viewer in the mind of the patient, makes him experience his confusion, his anger, his fear.

Hopkins remembers that when he read the script he felt something similar to what happened to him with “The Silence of the Lambs.” He knew instantly that he wanted to. “I met with Florian and said yes, but I had the filming of ‘The Two Papas’ pending (with Jonathan Pryce and the direction of Fernando Meirelles) and I was really worried that they would not wait for me, but they did,” he says. .

In the case of Hannibal Lecter, Hopkins recalls that when he began to read the script, he asked his agent if the deal was already closed. “He said no and I didn’t want to read any more until I knew, because he was the best character I could have ever dreamed of.”

He says that to keep his brain active he plays the piano, paints or memorizes poems by Dylan Thomas, but when it comes to talking about his merits as an actor or uncovering the secrets of some of the most legendary scenes of his career, he insists on subtracting importance.

“Acting does not require being a genius or a great intellectual effort, there will be people who are interested in making it look like that, but no,” he defends.

“In ‘The Silence of the Lambs’ Jodie came with a coffee in hand, we would stare at each other through a glass wall and blurt out the text. That’s it. We still meet sometimes to eat together and talk about how funny this is way of making a living. “

According to Zeller, Hopkins is an instinctive actor. “I am simply old”, replies the interpreter, “I have been active for many years so I know some tricks; the Stanislavski method is wonderful and I practiced it when I was young, but now it is more common sense than anything else”.

“When you get to 83, I don’t know if you’re smarter or stupider, but you certainly don’t waste your time thinking too much about yourself,” he adds. “When it comes to acting, I just do it and that’s it, and with an actress like Olivia, a wonderful director and a good script like this one, that’s the most important thing,” he adds in reference to “The father.”

However, at another point in the interview he confesses that when they were filming, between takes, he would stand in front of a mirror and repeat to himself: ‘This is just a game, okay? I am not suffering from dementia, I am acting, I am acting. ‘

“You have to be careful with the message that you transfer to your subconscious because it can be believed,” he explains. “I remind my brain of things, when it tells me that my memory is failing or that I am getting older, I say no, that I am still young and strong, and the brain believes it, it really works.”

He also does not practice looking back too much. “My past seems like a dream to me, I feel like I can’t be responsible for anything, I don’t know why I became an actor, I don’t remember, I tend to think that everything has been a solipsism, that my parents never existed, they are only memories” .

His father died more than 40 years ago from heart disease. “He was in the hospital and I knew he was dying, that he was not going to leave, but suddenly you see yourself making strange promises; I told him that when he left I would take him on a car trip from New York to Los Angeles and a few days later I went to see him and he was sitting there with a road map of the United States, so happy. “

“It is painful when you know that they are dying but I suppose the only thing you can do is reassure them and let them know that you are there,” he says.

His mother passed away years later. “My wife Stella was with her and to reassure her she told her that when she got to the other side she would meet her husband again. My mother, who was not very religious, replied: ‘Will it be very expensive?’ He had a great sense of humor at the end. ”

While computers have long been used to generate data for mathematicians, the task of identifying interesting patterns has relied mainly on the intuition of the mathematicians themselves. However, it?s now possible to generate more data than any mathematician can reasonably expect to study in a lifetime. Which is where machine learning comes in.

A paper, published today in Nature, describes how DeepMind was set the task of discerning patterns and connections in the fields of knot theory and representation theory. To the surprise of the mathematicians, new connections were suggested; the mathematicians were then able to examine these connections and prove the conjecture suggested by the AI. These results suggest that machine learning can complement mathematical research, guiding intuition about a problem.

Using the patterns identified by machine learning, mathematicians from the University of Oxford discovered a surprising connection between algebraic and geometric invariants of knots, establishing a completely new theorem in the field. The University of Sydney, meanwhile, used the connections made by the AI to bring them close to proving an old conjecture about Kazhdan-Lusztig polynomials, which has been unsolved for 40 years.

Professor Andras Juhasz, of the Mathematical Institute at the University of Oxford and co-author on the paper, said: ?Pure mathematicians work by formulating conjectures and proving these, resulting in theorems. But where do the conjectures come from?

?We have demonstrated that, when guided by mathematical intuition, machine learning provides a powerful framework that can uncover interesting and provable conjectures in areas where a large amount of data is available, or where the objects are too large to study with classical methods.?

Professor Marc Lackeby, of the Mathematical Institute at the University of Oxford and co-author, said: ?It has been fascinating to use machine learning to discover new and unexpected connections between different areas of mathematics. I believe that the work that we have done in Oxford and in Sydney in collaboration with DeepMind demonstrates that machine learning can be a genuinely useful tool in mathematical research.?

Professor Geordie Williamson, Professor of Mathematics at the University of Sydney and director of the Sydney Mathematical Research Institute and co-author, said: ?AI is an extraordinary tool. This work is one of the first times it has demonstrated its usefulness for pure mathematicians, like me.

Older women are among the most susceptible to developing UTIs, an infection of the bladder and urethra that causes urinary urgency and pain. UTIs also can cause delirium in older people, resulting in a sharp decline in mental abilities that triggers disoriented thinking.

?Up to one-third of elderly patients hospitalized with UTIs can experience some degree of confusion and reduced awareness of their surroundings,? said Shouri Lahiri, MD, director of the Neurosciences Critical Care Unit and Neurocritical Care Research at Cedars-Sinai and senior author of the study. ?Delirium affects millions of patients a year in the U.S., contributing to longer hospital stays, long-term cognitive problems and increased mortality. Delirium can be a tipping point from which patients never fully recover. This is well established. What is less well established is why this is happening.?

To better understand the specific biological mechanisms behind UTI-associated delirium, Lahiri and colleagues observed laboratory mice with and without UTIs in specially designed mazes. In an arena where the animals could move about freely, uninfected mice spent more time in the center of the chamber. Those with UTIs huddled in the periphery, suggesting they had higher levels of anxiety, a common symptom of delirium.

In a Y-shaped maze with three arms to explore, uninfected mice tended to explore all three arms, while mice with UTIs kept returning to the same one, suggesting a lapse in short-term memory, another feature of delirium.

The investigators also observed structural changes in the brains of mice with UTIs.

In a previous study led by Lahiri, published in February in the American Journal of Respiratory Cell and Molecular Biology, investigators found a connection between ventilator-induced lung injury and delirium. Lahiri and colleagues theorized that in both cases this was because of the reaction of IL-6, which helps regulate immune response, to the lung injury or the UTI.

?Occasionally, when the response of IL-6 is excessive, our research indicates that there can be brain injury,? Lahiri said. ?IL-6 induces changes within the neurons that our studies connected with delirium-like behavior. This is the first time this type of structural and functional change has been demonstrated. We?ve now shown two distinct models of this connection, one non-infectious and one infectious.?

In the current study, when investigators treated some of the infected mice with antibodies that blocked the effects of IL-6, the delirium-like behavior of those animals resolved. ?Treatment with anti-IL-6 antibody in the UTI group normalized all the brain changes, both structural and functional,? Lahiri said. ?A wealth of studies have shown a link between IL-6 and delirium, but only this study and our previous study have shown that IL-6 may play a direct pathological role in delirium.?

If symptoms are treated early, he added, full recovery is possible, and the next step is to design clinical trials with anti-IL-6 antibodies as a treatment for patients with UTI-induced delirium.

?Dr. Lahiri and his team have built a research framework with implications far beyond this study,? said Nancy Sicotte, MD, chair of the Department of Neurology who holds the Women?s Guild Distinguished Chair in Neurology at Cedars-Sinai. ?Building on these efforts could improve outcomes for many of our patients.?

Lahiri believes the model he and co-investigators created could be used to study delirium in other conditions associated with increased incidence of UTI. These include those with pre-existing neurologic disorders like Alzheimer?s disease, Parkinson?s disease, stroke and multiple sclerosis where delirium can be hard to distinguish from the underlying disease but is important to treat.

?A huge population stands to benefit from these investigations,? he said. ?We?re looking to apply this model to these other systemic disease conditions and states where the brain dysfunction caused is potentially reversible.?
